Next, we examined the effect of finasteride on the hair growth of mouse hair follicles treated with finasteride or finasteride alone or with finasteride and finasteride alone. In addition, we examined the effects of finasteride on male pattern baldness by measuring the hair follicle counts and the hair growth in a rat model of male pattern baldness.
Rats were purchased from the National Foundation for Hair Restoration (New Jersey, USA) and were housed under a 12-h light/dark cycle. Male and mouse female rats were housed in a polyethylene catroom (Neuro-Medica, Inc., USA) with a 12-h light/dark cycle. All animal procedures were in accordance with the Guide for the Care and Use of Laboratory Animals, and conducted in accordance with the Declaration of Helsinki. Male and mouse female rats were used in this study for the experiment. Male rats were weighed and given a 1% castor oil suspension intraperitoneally (i.p.) daily for 6 weeks (total length of the experiment was 9 weeks). Rats were sacrificed by cervical dislocation on the third week of the experiment, and hair follicles were removed using a scalpel and fixed in 10% formalin. After washing the follicles with PBS, the hair follicles were dehydrated in phosphate buffer saline (PBS) for 10 min, and the scalp was embedded in paraffin. The hair follicles were then excised, and the hair follicle area was determined using a light microscope. The hair growth was measured using the ImageJ®-Pro® (National Institutes of Health, USA).
